From the start of the game, you only can play as Joe on Kids or Adults mode. Here are the various rewards you get for your accomplishments in the game:

Unlimited VFX is without a doubt the hardest thing to unlock in Viewtiful Joe. To go about unlocking it is generally referred to as the "Rainbow-V Challenge." To get a Rainbow V rank for a level, you cannot have even one task with a regular V ranking, or else you will fail. Soft resets (hold B+X+Start for the Gamecube) are very useful, and patience should be kept in good supply.
To use your Unlimited VFX, start a new game. When selecting your character, hold down Z on the Gamecube before selecting them. You should see "Super" over their name, and the text should billow faster. Then just go along until you get VFX; you'll find that the meter never goes down unless you do a Slow-Dodge.
All characters have the same cutscenes as Joe's story with certain models replaced. All dialogue is dialogue from Joe's story played backwards and with various tone alterations.
Sylvia runs twice as fast and has a slightly higher jump than Joe, making her mobility the core of her play style. She takes double damage, also reiterating agility versus power, and she does not deal as much damage as Joe. Her combos on the basic enemies last around 12 hits, increasing her potential for V's, but it becomes a challenge when dealing with multiple enemies.
Alastor uses "Devil Trigger" to enter his Henshin form by pressing Z to toggle the Devil Trigger. When he is in Henshin mode, his VFX constantly drains. However, this allows him certain advantages over the other characters, most notably that he can use Mach Speed for much longer periods of time. When in Henshin mode, you can control the rate he falls with the analog stick; by default, he floats along fairly slowly but can be made to fall faster than the other characters. In normal mode, he will strike enemies with his sword after punching 4 times in a combo. He can also double jump in normal mode; when he does, he yells "Air Hike!," a reference to Devil May Cry. Just about everything about this character is a reference to Devil May Cry.
Captain Blue cannot see skulls. That's no big deal if you completed Ultra-V Rated, though. He also doesn't double-jump; he has one big jump, and holding jump after doing that will make him float in midair. He doesn't lose altitude at all while floating and can float for indefinite periods of time. One can perform several minor sequence breaks this way. He is also the strongest character in the game.
Dante can attack with swords and two handguns. The finishing sword thrust of his ground attack combo, when combined with VFX powers Slow and Zoom, does massive damage actually enabling a one-hit kill on Fire Leo on Adults difficulty.
