Lloyd Irving grew up just outside Iselia, raised by his Dwarven foster father, Dirk. While not the brightest student in Raine Sage's classroom, he makes up for it with a strong sense of right and wrong and a fierce determination. At times, that determination can turn into sheer stubbornness, and this behavior occasionally bothers other members of the group. Lloyd is a quick fighter who uses two swords, due to the belief that two swords must deal twice as much damage as a single sword. While he is never able to use any true magic, his special attacks are still quite powerful.

Genis Sage is the child prodigy of Iselia and best friend of Lloyd Irving, as well as younger brother of Raine Sage. He frequently belittles Lloyd, in a friendly manner, and pokes fun at Lloyd's lesser intellect and inability to remain awake and alert in Raine's class. As the magic user of the group, Genis has the potential to deal a tremendous amount of damage, but his ability to receive damage is very limited, and his concentration can be easily broken by an enemy attack. His preferred weapon is the Kendama, but he rarely uses it to attack and instead uses the various Kendama to enhance his magic. One skit showed Genis trying to teach other members of the party to use the Kendama with unsuccessful results.

Raine Sage is the local teacher for the small town of Iselia and has lived there since her younger brother Genis was a small child. She has vague memories of a time before Iselia, but has trouble placing where those memories came from. It is in an attempt to place the memory of a particular ruin that has driven her to be passionate about archeology. Her passion borders on obsessiveness, a trait commented on by the other characters on numerous occasions. She is the healer of the group, and thus is not a skilled fighter and not capable of dealing or receiving much damage. Nonetheless, she is a great addition to any battle party since she can heal allies' health, resurrect fallen allies and grant status enhancements in combat, as well as remove status ailments.

Collette Brunel is the most recent Chosen of Mana, and as such has a destiny to unlock the four elemental seals and rejuvenate the flow of mana in Sylvarant. She is also very clumsy and apologetic, the former trait being so prominent, the first optional title to be gained in the game is her 'Klutz' title, which can be gained within minutes of initially gaining control of Lloyd at the start of the game. She uses dual chakram in combat and has a blurred role, being able to deal moderate damage through both melee based special skills and the angelic based magic she gains during the course of the journey.

Kratos Aurion is a mercenary who encounters the group outside the Iselia temple and comes to Lloyd, Collette and Genis' aid during a difficult fight. His past is unclear and his given motives are that as a mercenary, he must take on work to earn money, and protecting Collette, the Chosen, on her journey of regeneration is just as good a job as any. He frequently gives harsh advice to Lloyd on his sword technique in the early stages of the journey. Kratos is both capable with a sword and with some minor magic, notably First Aid, making him a versatile member of the group. While he can be used as a light healer, his primary skill remains as a front-line fighter.

Sheena Fujibayashi is a resilient, and clumsy, assassin who attempts to kill Collette during the course of the journey. Her past and motives are initially unknown, but once she reveals where she is from and why she is after Collette, the nature of the journey comes under question and Sheena joins the group in their new goal. She is the only summoner of the group, and is able to form pacts with the various summons the group encounters and call forth their power in battle. Besides her summoning ability, she uses magical cards in combat to deal damage and place status effects on the enemy.

Zelos Wilder is a flamboyant playboy who lives a life of luxury as a Chosen of Mana. Once he encounters the group and discovers what they are up to, he is forced to rapidly adjust to a whole new understanding of the world, and his role as the Chosen. He fights with a very similar style to Kratos Aurion, and is even able to equip the same gear.

Presea Combatir appears to be a young girl with a stoic, even robotic demeanor. While she is small, she possesses incredible strength, able to pick up and move objects that not even the adult members of the team can handle. She wields an axe with ease and can lay down a large amount of damage, even though she attacks more slowly than the nimble swordsmen of the group. Genis is attracted to her at first glance, but she doesn't return the sentiment and barely notices his compliments.

Regal Bryant is a shackled prisoner who is tasked with eliminating the main group. For a convict, he is shockingly well mannered and articulate, leading the group to question Regal's life before being imprisoned, along with the nature of his crimes. He refuses to use his hands as weapons and fights purely with his feet, while his wrists are bound together. As a fighter, he is able to both take and receive damage with ease.
