As Zombie Mushrooms spawn, they will slowly make their way to the bottom floor. After enough enemies are dead, the top floors will be empty while the bottom is overrun. If you're worried about taking damage, spend the entire time on the bottom floor so that enemies don't congregate in groups too large for you or your party to handle.
