Assassin's Creed: Brotherhood keeps its sandbox style gameplay in an open world based on the city of Rome. Like the previous installments, the gameplay's core mechanics are based on [[wp:Parkour|Parkour]] movements, crowd-blending stealth, assassinations and melee fighting system. However, for the first time in the series, the game offers a multiplayer mode alongside 15 hours of single player campaign.

According to Ubisoft, the game is going to offer over 15 hours of [[single player]] gameplay set in the city of Rome with the possibility to recruit and train young Assassins that can be deployed across the city, or call upon to aid Ezio in his quests. Also in the quests, Ezio will meet real historical characters such as Leonardo da Vinci, Niccolò Machiavelli and Caterina Sforza with which he can collaborate, and also by helping out citizens of Rome and convincing them to join the cause will unlock extra factions and missions. As in weaponry, the game will have old and new weapons to use on against enemies like poison darts, parachutes, double hidden blades, hidden guns, and an advanced flying machine.
As a Master Assassin, Ezio must reorganize his Brotherhood in Rome to fight against the Templars. The player can recruit new members by completing missions around the city, send them to assignments around Europe or call them for support during missions (if they are not already occupied). Tasking the novice Assassins is essential to make them gain experience, and the player is able to customize their appearance, skills and weapon training by spending the skill points they've earned.

Multiplayer mode is new to the franchise and will allow the player to choose from multiple authentic character classes, each with their own signature weapons and killing moves to battle on maps and a wide variety of multiplayer modes.
The multiplayer experience is available on all platforms. The multiplayer beta was exclusive to PlayStation 3 users and began on October 4, 2010, lasting just under two weeks. In multiplayer, the players are Templars in training at the Abstergo facility. They use the animi (plural for animus) seen at the beginning of Assassin's Creed II to access memories of Assassins and then to acquire their skills using the "bleeding effect". There are multiple modes and maps in the multiplayer as well as its own skill and rank advancement systems separate from the single player campaign.
